Principle and Applications
This kit consists of an enzyme-linked immunosorbent assay (ELISA) plate pre-coated with foot-and-mouth disease type O antibodies, antibody working solution, antigen-enzyme conjugate, and other accompanying reagents. It employs the principle of competitive ELISA to detect foot-and-mouth disease type O antibodies in serum samples from cattle, sheep, and pigs. This kit is suitable for detecting foot-and-mouth disease type O antibodies in the serum of cattle, sheep, and pigs, and can be used for evaluating the efficacy of foot-and-mouth disease type O vaccination as well as for auxiliary diagnosis.
Composition of the kit
Sample Preparation
In the serum dilution plate, dilute the serum to be tested 5-fold using the sample diluent (for example, add 25 μL of the serum to be tested to 100 μL of sample diluent). The positive control and negative control should also be diluted 5-fold.
Testing method
1. Before use, place the kit at room temperature for at least 30 minutes to allow it to return to room temperature (20–25℃).
2. Take the required number of ELISA strip wells. Set up 1 well for the blank control and 2 wells each for the negative and positive controls. Seal any unused strips promptly and store them at 2–8°C. Then wash the plates twice using the pre-diluted washing solution.
3. Add 50 μL of sample diluent to the blank control well; add 50 μL of negative and positive controls respectively to the negative and positive control wells. Then, add 50 μL of enzyme conjugate to each well, gently shake to mix thoroughly, and incubate at 37℃ for 30 minutes.
4. Discard the reaction liquid, add 200 μL of washing solution to each well, without allowing it to sit. Spin off the washing solution from each well, repeat the washing process five times, and finally blot dry on clean absorbent paper.
5. Add 100 μL of substrate solution to each well and develop the color at room temperature in the dark for 15 minutes.
7. Add 50 μL of stop solution to each well, mix thoroughly, and within 10 minutes, measure the absorbance (OD value) of each well at a wavelength of 450 nm (a reference wavelength of 630 nm may be used). If no reference wavelength is available, zero the instrument using the blank well (i.e., subtract the absorbance of the blank well from the absorbance of each well).
