【Characteristics】 Loose clumps that easily detach from the bottle walls and rapidly dissolve upon addition of a diluent.
[Function and Use] Used for the Brucella agglutination test on plate and the Brucella tube agglutination test, for the diagnosis of brucellosis.
[Usage and Judgment] This product is a lyophilized preparation that must be fully dissolved in 1.0 ml of sterile distilled water before performing the Brucella agglutination test on tiger-red plates and the Brucella tube agglutination test.
(1) For the Brucella Bengal Rose Plate Agglutination Test
Perform a plate agglutination test by mixing 0.03 ml of Brucella antigen with an equal volume of Brucella-positive serum. Agglutination can be visually observed within 4 minutes.
(2) For the Brucella tube agglutination test
Dilute the Brucella agglutination test antigen with 0.5% phenol-saline solution at a ratio of 1:20. Dilute the Brucella-positive serum at ratios of 1:12.5, 1:25, 1:50, 1:100, and 1:200. Add 0.5 ml of each antigen and serum to separate test tubes, incubate at 37℃ for 24 hours, and then observe the agglutination results. The results showed that the antigen elicited an agglutination titer of “++++” when reacted with Brucella-positive serum at all dilutions.
[Precautions]
(1) Use immediately after opening.
(2) The antigen and serum should be allowed to equilibrate at room temperature for 30 to 60 minutes before the assay is performed.
