Method for Purifying Bovine Tuberculin

Mar 07,2022

Method for purifying bovine tuberculin: When gas is stimulated by Mycobacterium tuberculosis, lymphocytes transform into lymphoblasts and proliferate, differentiating into sensitized lymphocytes that become sensitized to the gas. Subsequently, upon subsequent contact with purified bovine tuberculin, these sensitized lymphocytes—on the one hand, release various lymphokines, and on the other hand, elicit a specific cellular immune response in the body. This type of reaction is characterized by mononuclear cell infiltration, macrophage release of lysosomes (hydrolytic enzymes), and tissue lesions caused by lymphocyte-mediated reactions.

After being stimulated by Mycobacterium tuberculosis, lymphocytes differentiate into lymphoblasts and proliferate, giving rise to sensitized lymphocytes that become sensitized to the gas. Subsequently, when these sensitized lymphocytes come into contact again with purified bovine tuberculin, they release various lymphokines on the one hand and elicit a specific cellular immune response in the body on the other. This type of reaction is characterized by mononuclear cell infiltration, macrophages releasing lysosomes (hydrolytic enzymes), and tissue lesions caused by lymphocyte-mediated reactions.

Injection Site and Preoperative Preparation: Shave the middle one-third of the neck (or shave the day before). Within 3 months, purified bovine tuberculin can also be injected into the scapular region, with a diameter of approximately 10 centimeters. Measure the thickness of the skinfold at the center of the injection site using calipers and record the measurement.

If the surgical site is changed, you must select a different site or proceed from the other side.

Purified bovine tuberculin injection dosage: Administer 10,000 international units intravenously in the bloodstream. The purified bovine tuberculin will soon be diluted to a concentration of 100,000 international units per liter; then inject 0.1 milliliter into the bloodstream. If using a 2.5-milliliter syringe, you should also inject an equal volume of saline solution subcutaneously. For intramuscular injection, administer 0.2 milliliter. Once the frozen bacterial suspension has been diluted, it must be used up within the same day.

Purify bovine tuberculin Injection method: First, disinfect the injection site with 75% alcohol. Then, inject a measured dose of small, pure tuberculin into the bloodstream. If you have any doubts about the injection procedure, you should select a separate or specially targeted site that is not within 15 centimeters, where slight foam may appear.

Number of injections and observation of reactions: Assess 72 hours after intradermal injection, carefully observing the local area for inflammatory reactions such as heat, pain, or swelling. Use calipers to measure the thickness of skin wrinkles and record the findings. If a reaction is suspected in one animal, repeat the same procedure on the other side using the same strain. Administer a second dose via intradermal injection and observe the reaction again 72 hours later. If possible, also evaluate the reactions in the vocal cords and other areas of suspicion.

Purified bovine tuberculin, when purified from normal animals, does not elicit any obvious local inflammatory response. In contrast, when injected into animals sensitized to tuberculosis, a delayed-type hypersensitivity reaction occurs. Within a few hours after injection, no macroscopic or histological changes are visible at the injection site; however, subsequently, vasodilation and increased transparency develop, accompanied by erythema, edema, swelling, and induration. Histological examination reveals that this lesion differs from a typical acute inflammatory response: the infiltrating cells are predominantly mononuclear cells—namely, macrophages and lymphocytes—and transient neutrophil accumulation may also occur in the early stages of the reaction. The intensity of the reaction reaches its peak between 24 and 72 hours post-injection and gradually subsides thereafter. This is a very severe reaction that can lead to necrosis at the injection site.
